The research aims to study the comparison of protease activity of three kind species of frangipani plant that is Plumeria rubra L, Plumria obtusa L and Plumeria pudica Jacq which mostly cultivated in Bali region especially in Badung regency. The assay of protease activity was based on spectrofotometric of Kurnitz modified Chanda method using casein as substrat. The result of protease activity assay of three kinds species of plumeria plant showed that species of Plumeria rubra L. had the highest protease activity which was 0.0398 ± 0.00034 U/mL, followed by Plumeria pudica Jacq. species 0.0371 ± 0.00019 U/mL, and was the lowest which protease activity of Plumeria obtusa L. species 0.0365 ± 0.00032 U/mL. The species of Plumeria rubra L. that was the most common plumeria species and cultivated in Bali region, therefore was potential as an alternative source of protease.
