Indonesia is a developing country with high incidence of diarrhea. E.coli is major cause of diarrhea in developing country. This gram-negative bacteria is capable to contaminate food and drinks. Jamu beras kencur is often consumed as a health drink, lack of supervision and hygiene of the manufacturing process, make it possible for E.coli to contaminate that health drink. This study aims to determine the contamination and serotype of the E.coli bacteria in jamu beras kencur sold by jamu gendong seller in Denpasar. Design of the study is descriptive observational with exploratory design. Sampling process was done by non-probability sampling method, that is consecutive sampling, in which ten of jamu beras kencur samples obtained from jamu gendong seller in four districts in Denpasar. Jamu specimens than mixed with Triptic Soy Broth (TSB) and inoculating on MacConkey media and Eosin Methylene Blue Agar (EMBA). Colonies that obtained then examined with gram staining and serotype identification using Sorbitol-MacConkey Agar (SMAC). Ten positive samples showed the presence of bacteria growing in TSB. On MacConkey medium and after subculture, six samples suspected E.coli positive. In gram stain process, show that ten samples obtained showed gram-negative bacillus bacteria. In EMBA media show that four samples suspected E.coli positive. The serotype identification show negative result from four samples that obtained before. The final result, four of ten (40%) samples suspected positive contaminated with E.coli, whereas O157 serotype of E.coli not found in the ten samples. Further research in needed regarding jamu beras kencur with larger sample.
