IDENTIFICATION OF Escherichia coli serotype O157 BACTERIA WITH SORBITOL MACCONKEY AGAR (SMAC) IN YOUNG COCONUT DRINK FROM STREET VENDORS IN THE RENON REGION AT SOUTH DENPASAR

Diarrheal disease is still a global problem with the high degree of morbidity and mortality in many countries, especially in developing countries . Escherichia coli serotype O157 is one of the most significant of E. coli cause infection in the human digestive tract. In humans, Escherichia coli O157 causes hemorrhagic colitis, hemolytic uremic syndrome (HUS) and thrombocytopenia purpura (TPP). E. coli O157 infection that pathogen in a human has the characteristic of verotoxigenic that caused 16,000 cases of the disease through food (Food Borne Diseases) and 900 people die each year in the US. Renon region located in the Denpasar city found many street vendors selling young coconut drink. The purpose of this research is to identify and determine how many bacteria Escherichia coli serotype O157 contaminating young coconut drink from a sample of street vendors in the Renon Region, South Denpasar. Type of research is descriptive observational study design explorative . In 10 samples that use non-probability/nonrandom sampling method namely quota sampling and analyzed at the Laboratory of Microbiology using TSB medium, MacConkey, gram staining, EMBA, SMAC and Singlepath® E. coli O157. Five of the 10 samples contaminated by bacteria Escherichia coli shown with metallic green colonies on media EMBA. In the SMAC test, one of the five samples contaminated by bacteria Escherichia coli colonies are colorless (colorless) showing the alleged contamination of bacteria Escherichia coli serotype O157. Confirmation test using rapid test with Singlepath® E. coli O157 showed a negative result (-), formed a red line in the control (C) but not formed a red line on the test (T).
