EFFECT OF NEWCASTLE DISEASE VIRUS VIRULENT ISOLATES TO HISTOPATHOLOGIC AND CHICKENS EMBRYO WEIGHT

Virulent Newcastle Disease Virus (NDV) cause interference to the nervous system, digestive system and respiratory system for poultry. It infected in the chickenís embryo caused growth disorders and died. This research conducted to determine the development of the chickenís embryo and histopathological lesions appearance of the brain, lungs, intestines, and bursa of fabricius post inoculated with isolates APMV type -1 Badung - 2/AK/2014. This research used 18 Chickenís Embryo Eggs (CEE) day 11 were divided into the two treatment groups, A and B, every groups had nine CEE. Group A inoculated with Phosphate Buffer Saline and group B inoculated with NDV isolates Badung - 2/AK/2014. Three days after inoculated, the allantoic fluid were taking to tested with hemagglutination and hemagglutination inhibition test. That chickenís embryos removed from the eggs shell to evaluated the weight of embryos and result analyzed by independent samples T-test. After the chickenís embryo weight, necropsied the brain, lungs, intestines and bursa of fabricius. These organs inserted into Neutral Buffer Formalin (NBF) 10% subsequently used for making preparations with staining technique Hematoxilin Eosin (HE). Histopathological appearance observed under a microscope and the histopathological changes were presented descriptively.The result showed that NDV virulent isolate caused growth disorders such as weight loss and damage histopathological lesion, vasculitis in cerebrum, inflammatory cells infiltration, necrosis, hemorrhage and limfositolis in the others organs.
